Lentivirus is a class of retroviruses that includes human immunodeficiency virus (HIV), simian immunodeficiency virus (SIV), bovine immunodeficiency virus (BIV), feline immunodeficiency virus (FIV), puma lentivirus, and equine infectious anemia virus (EIAV). Among these viruses, HIV is widely used and has become the standard for lentiviral vectors (LV) for delivering genetic material (DNA or RNA) into target cells. HIV-1-derived lentivirus (LV) is known for its efficient and stable transduction in both dividing and non-dividing cells. It utilizes viral integrase to integrate the desired transgene into the target cell genome. In addition, LV also has the advantages of broad cell tropism and targeting specific cell types through pseudoviruses.
LV can be stably integrated into the target cell genome as a provirus. The third-generation lentiviral packaging system is safer and suitable for production in HEK293T cell lines. Some other studies have reported that the use of SV40 T antigen can increase vector yield. The vectors obtained 48 hours and 72 hours after transfection can be harvested from the supernatant. To produce fragment-free lentivirus, further purification and concentration should be performed.
