Description This product is a type of oncolytic adenovirus (OAd) that is derived from human adenovirus serotype 5 (hAd5). This OAd contains E1A and E1B in which a 24 bp in the conserved region 2 (CR2) of E1A is deleted. It also contains GFP reporter gene expression cassette, and the E3 region in this OAd is completely deleted. The CR2 (Delta-24) mutation enables this OAd to infect tumor cells with mutations in the Rb/p16 pathway and lead to cell lysis.
Background The use of oncolytic adenoviruses (OAds) is a promising approach in cancer therapy as they can be engineered to specifically replicate in tumor cells, while leaving healthy cells unharmed. The replication of OAds in tumor cells eventually leads to lysis and death of tumor cells. Additionally, they can be combined with other treatments, such as chemotherapy or radiation therapy, to enhance their effectiveness. Clinical trials have shown some success in using oncolytic adenoviruses to treat various types of cancer, including head and neck cancer, melanoma, and bladder cancer etc.

RCA Assays Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources.
PFU Titering All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells.
