Purity AAV purity is critical for ensuring the safety and efficacy of AAV-based applications.AAV capsids are composed of three main protein components, known as viral proteins: VP1, VP2, and VP3. These proteins play a critical role in the structure and functionality of the AAV capsid. Monitoring the VP1, VP2, and VP3 content in AAV preparations is essential for quality control in AAV production. Our AAV particles are tested for showing three clear bands of VP1, VP2 VP3 by SDS-PAGE.
Sterility The AAV virus samples are inoculated into the cell culture medium for about 5 days to detect bacterial and fungal growth.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of AAV to deliver genetic material into target cells or tissues, and assess gene expression and functional activities.

Adeno-associated virus (AAV) was first discovered in the 1960s as a contaminant of adenovirus stocks. Gene therapy based on recombinant AAV (rAAV) vectors has attracted increasing attention worldwide. The most widely studied serotype of AAV is type 2 (AAV2), which is the prototype of the AAV family. The AAV2 genome is a ssDNA of approximately 4.7 kb. It is flanked by 145 nt of inverted terminal repeats (ITRs) that form a T-shaped base-paired hairpin structure and contain cis-elements required for viral genome replication, encapsidation, and integration into and rescue from host chromosomal DNA. The AAV2 genome contains two ORFs. rep encodes four nonstructural proteins required for replication (Rep78, Rep68, Rep52, and Rep40), while cap encodes three structural proteins (VP1, VP2, and VP3) that constitute the viral capsid. Three viral promoters have been identified based on their relative map positions within the viral genome: p5, p19, and p40. Efficient AAV2 infection requires helper functions, which can be provided by a co-infected helper virus or a DNA damaging agent. Helper viruses include adenovirus, herpes simplex virus, vaccinia virus, cytomegalovirus, varicella-zoster virus, and human papillomavirus.
