Monitoring the VP1, VP2, and VP3 content in AAV preparations is essential for quality control in AAV production. Our AAV particles are tested for showing three clear bands of VP1, VP2 VP3 by SDS-PAGE.
Sterility The AAV virus samples are inoculated into the cell culture medium for about 5 days to detect bacterial and fungal growth.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of AAV to deliver genetic material into target cells or tissues, and assess gene expression and functional activities.

Adeno-associated viral (AAV) vectors are small, non-enveloped, non-pathogenic, single-stranded DNA parvoviruses that infect a variety of cell types, including many types of neurons and glial cells. Thirteen naturally occurring AAV serotypes of human and simian origin have been reported, which are recognized by different cell surface receptors and have different cellular affinities. For example, astrocytes are the predominant cell type transduced by AAV serotype 8, whereas AAV serotype 9 (AAV9) primarily transduces neurons. Deverman et al. reported AAV-PHP.B (hereinafter referred to as PHP.B), which is a capsid variant of AAV9 with 7 amino acids inserted, showing an extremely high ability to penetrate the mouse blood-brain barrier (BBB). . Intravenous infusion of PHP.B effectively transduces the entire brain. In addition, the research team also modified AAV-PHP.eB (hereinafter referred to as PHP.eB), which differs from PHP.B by only 2 amino acids near the heptamer insertion site. After intravenous injection of PHP.eB, the transduction effect on cerebral cortex neurons was significantly enhanced, and the transduction efficiency was higher than that of PHP.B.
