SDC1 or Syndecan-1 is a transmembrane heparan sulfate proteoglycan that plays a key role in cell communication, signaling pathways, and cell-extracellular matrix interactions. Syndecan-1 is primarily expressed on the surface of epithelial cells, particularly in tissues such as the skin, breast, and gastrointestinal tract, as well as in certain hematopoietic cells. One of the main functions of SDC1 is its involvement in cell adhesion. As a coreceptor, it binds to a variety of ligands, including growth factors, chemokines, and extracellular matrix components such as fibronectin and collagen. Through these interactions, SDC1 mediates processes such as cell proliferation, migration, and differentiation, significantly affecting tissue morphogenesis and repair. In addition to its biological functions in normal physiology, SDC1 has been extensively studied in pathological contexts. Its altered expression has been associated with a variety of diseases, including various types of cancer, where its expression levels are often correlated with tumor progression, metastasis, and patient prognosis. For example, elevated levels of SDC1 are commonly observed in multiple myeloma, which not only provides a diagnostic biomarker but also a potential therapeutic target. Targeting SDC1 via antibodies or small molecules has shown promise in preclinical studies, aiming to disrupt its interaction with key growth factors or modulate its shedding from the cell surface.
Cancer Research: The Human SDC1 Knockout Cell Line-HeLa is frequently used in cancer research to investigate the role of syndecan-1 (SDC1) in cell proliferation, migration, and tumor growth. Cell Adhesion and Migration Studies: SDC1 is known to play a significant role in cell adhesion and migration. Using the knockout HeLa cell line allows scientists to explore how the absence of SDC1 affects these cellular processes, providing insights into mechanisms underlying wound healing, tissue regeneration, and cancer cell dissemination. Drug Screening and Development: This specific cell line is instrumental in high-throughput drug screening to identify compounds that can inhibit or modulate pathways influenced by SDC1. This aids in the discovery of new drugs aimed at treating cancers or other diseases where SDC1 is implicated. Signal Transduction Pathways: Researchers utilize the SDC1 knockout HeLa cell line to dissect the intricate signal transduction pathways mediated by SDC1. Understanding these pathways is vital for uncovering new molecular targets for therapeutic intervention in diseases like cancer and fibrosis. Glycobiology Studies: Syndecan-1 is a key player in glycobiology, interacting with various growth factors and extracellular matrix components through its glycosaminoglycan chains. The knockout cell line offers a model to study the biological functions of these glycan interactions and their implications in health and disease.
Customer Q&As
What is the recommended growth medium? Does it require antibiotic selection?

A: DMEM supplemented with 10% fetal bovine serum.
It is not required to add the selection antibiotics when culturing the KO cells.

How is the knockout cell line validated?

A: The knockout cell product is validated by PCR amplification and Sanger Sequencing to confirm the mutation at the genomic level. Please find the detailed mutation info in the datasheet.

Is the product a single clonal cell or mixed cell pool?

A: Single clonal cell.

Can I confirm gene knockout by RT-qPCR?

A: No. This knockout cell product is generated using the CRISPR/Cas9 system to induce small insertions or deletions (indels) resulting in frameshift mutations. Although these frameshift mutations typically disrupt the coding gene, there is a possibility that the non-functional transcript may still be transcribed. Consequently, this could potentially yield misleading results when analyzed by RT-qPCR.

How can I store the cell product?

A: The cell line should be stored in liquid nitrogen for long-term preservation.

Is it possible to get multiple knockout clones for my GOI?

A: For most cases, we often keep at least 2 clones with different frameshift mutations. Please feel free to contact us to check if there are additional available clones.
