Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

scFv(GPC3)-41BB-CD3zeta CAR-T lentivirus is a gene delivery vector designed to robustly genetically modify primary human T lymphocytes to express a second-generation chimeric antigen receptor (CAR) specifically targeting Glypican-3. One of the main advantages of using lentiviral vector systems is their ability to achieve stable, long-term genome integration, ensuring that the CAR transgene is delivered to all progeny cells during T cell expansion. This is crucial for achieving sustained therapeutic effects in adoptive cell therapy. The vector exhibits high transduction efficiency even in quiescent or slowly dividing primary T cells, a characteristic that significantly outperforms traditional retroviral vectors. Structurally, the second-generation CAR encoded by this virus contains a high-affinity single-stranded variable region fragment (scFv) derived from an anti-GPC3 antibody, optimized for precise antigen recognition. It is linked to a 4-1BB (CD137) co-stimulatory domain, which plays a key role in enhancing the metabolic adaptability and long-term survival of T cells in the host.

In terms of applications, scFv(GPC3)-41BB-CD3zeta CAR-T lentivirus is an indispensable tool in oncology and immunotherapy research, particularly suitable for treating hepatocellular carcinoma (HCC) and other GPC3-positive malignancies such as yolk sac tumors, squamous cell lung cancer, and embryonal carcinoma. Because GPC3 is overexpressed on the surface of these cancer cells but almost not expressed in healthy adult tissues, it is an ideal candidate target for CAR-T therapy with extremely low off-target toxicity. Researchers have used these lentiviral particles to construct standardized batches of CAR-T cells for various functional assays. In vitro, the system quantifies the cytotoxicity of modified T cells and detects the secretion of pro-inflammatory cytokines such as interferon-γ (IFN-γ) and interleukin-2 (IL-2) through co-culture experiments with GPC3-expressing target cells. Furthermore, by injecting the resulting CAR-T cells into tumor-bearing immunodeficient mice, scientists can study the cell biodistribution, their ability to infiltrate dense solid tumor stroma, and their efficacy in reducing tumor volume over time.
