Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv(GPC3)-CD28-OX40-CD3zeta CAR-T Lentivirus is a ready-to-use lentiviral vector that delivers a chimeric antigen receptor gene into human T cells. The construct encodes an anti-GPC3 single-chain variable fragment (scFv) as the extracellular antigen-binding domain, followed by the CD28 and OX40 costimulatory domains and the CD3zeta signaling domain in the intracellular region. GPC3, or glypican-3, is a cell surface proteoglycan that is highly expressed in hepatocellular carcinoma and several other solid tumors but is nearly absent from normal adult tissues, which makes it an attractive target. The vector is pseudotyped with the VSV-G envelope, allowing efficient transduction of primary T lymphocytes and T cell lines, and it integrates stably into the host genome through reverse transcription. This design combines the strong co-stimulation of CD28 with the OX40 domain, which is known to promote T cell survival, proliferation and memory formation, to support long-lasting antitumor activity in engineered T cells.

This lentivirus is mainly used to generate GPC3-specific CAR-T cells for research in solid tumor immunotherapy. Typical applications include ex vivo transduction of donor-derived T cells for in vitro cytotoxicity assays, cytokine release measurements and repeated antigen challenge studies that reveal T cell exhaustion and persistence. The product also serves preclinical evaluation in immunodeficient mouse models bearing GPC3-positive xenografts, where tumor growth inhibition and infiltration of engineered T cells can be assessed. Researchers also use it to compare different costimulatory configurations, to test scFv affinity and spacer design, and to establish stable cell lines or study GPC3-related biology. Beyond basic studies, it supports early-stage development of CAR-T therapies against hepatocellular carcinoma, hepatoblastoma, yolk sac tumor and other GPC3-positive malignancies, providing a practical tool for validating candidate constructs before moving into clinical manufacturing.
