Norovirus remains a major cause of epidemic acute gastroenteritis, with substantial global morbidity and mortality, yet no licensed vaccine is currently available. The researchers investigated avian immunoglobulin Y (IgY) antibodies as a cost-effective intervention against norovirus infection. Recombinant protruding (P) domain proteins from the capsid VP1 of multiple norovirus genotypes (two GII.4 variants and GII.6) were produced and combined into a single immunogen to immunize laying hens. The resulting yolk-derived IgY antibodies were characterized for their ability to inhibit binding of norovirus P particles to histo-blood group antigens and to neutralize viral replication in human intestinal enteroid (HIE) models. Protein expression and purification were performed using the GST fusion system, with subsequent thrombin digestion and anion exchange chromatography to ensure solubility and purity. The Creative Biogene plasmid vectors and recombinant protein systems enabled precise production of the P and S domain antigens, facilitating downstream evaluation of IgY efficacy.
