Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

EF1a-HPV16/E6(GFP, Puro) lentiviral particles represent a cutting-edge tool in gene manipulation and cell research. This genetically engineered lentivirus expresses the HPV16 E6 oncogene under the control of the EF1a promoter, ensuring stable and sustained expression in various mammalian cell types. The vector integrates a GFP reporter gene, allowing for real-time visualization and tracking of transduced cells via fluorescence microscopy or flow cytometry. Furthermore, the introduction of a puromycin resistance gene effectively screens and enriches stably transduced cells, significantly reducing experimental background noise. This lentiviral system exhibits high transduction efficiency, stable genome integration, and the ability to infect both dividing and non-dividing cells, surpassing traditional transfection methods. Its self-inactivation (SIN) design minimizes the risk of insertional mutations, improving biosafety, while the pseudotype envelope broadens tropism, making it promising for diverse applications.

This lentiviral vector is particularly important for cell immortalization research, as the HPV16 E6 protein plays a crucial role in disrupting the p53-mediated tumor suppressor pathway. E6 promotes cell proliferation and prolongs lifespan by degrading p53, thus facilitating the establishment of immortalized cell lines from primary tissues. Researchers can utilize these viral particles to mimic carcinogenesis, study the function of viral oncoproteins, and explore mechanisms of cellular senescence. GFP labeling simplifies the monitoring of transduction efficiency and cell lineage tracking, while puromycin screening ensures cell population homogeneity, resulting in reproducible results. Its applications encompass drug screening platforms, cancer biology research, and the construction of in vitro disease models. Furthermore, this system enables long-term functional studies of the roles of HPV16 E6 in DNA damage response, apoptosis regulation, and telomerase activation, providing new insights for therapeutic interventions.
