Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Customer Reviews

EF1a-hHOXA9 (RFP, Bla) lentiviral particles are cutting-edge tools in gene manipulation and cell engineering. These particles are designed to deliver the human HOXA9 gene under the efficient control of the EF1a promoter, ensuring stable and sustained expression in a variety of mammalian cell types. This lentiviral vector system offers numerous advantages, including high transduction efficiency, stable genome integration, and the ability to transduce both dividing and non-dividing cells. This makes it particularly important for primary cell lines or stem cells that are difficult to handle with other transfection methods. Furthermore, the introduction of the RFP reporter gene allows researchers to easily observe and track transduced cells using fluorescence microscopy or flow cytometry, enabling real-time monitoring of transduction efficiency and transgene expression. The blast fungicide resistance gene provides a powerful screening mechanism, allowing researchers to enrich successfully transduced cells and maintain stable cell populations under antibiotic stress. This dual-selection system improves experimental reliability by minimizing background noise from untransduced cells.

The primary application of EF1a-hHOXA9 (RFP, Bla) lentiviral particles is in cell immortalization. HOXA9 is a homeobox transcription factor whose overexpression induces durable proliferative capacity in normally senescent primary cells. This is particularly useful for establishing continuous cell lines from limited patient-derived samples, such as hematopoietic stem cells, fibroblasts, or epithelial cells, which can serve as a renewable resource for long-term research. In cancer research, immortalized cell lines expressing HOXA9 can be used to study oncogenic pathways, drug resistance mechanisms, and tumor microenvironment interactions. The RFP reporter gene further supports its application in cell migration assays, co-culture systems, and in vivo tracking in xenograft models. Beyond immortalization, these particles also play a crucial role in functional genomics research, exploring the role of HOXA9 in development, differentiation, and disease pathogenesis, including its significance in leukemia and solid tumors.
