Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The cAMP/PKA luciferase reporter lentivirus is an advanced research tool. Its lentiviral particles are genetically engineered to carry a luciferase reporter gene regulated by a minimal promoter and a cyclic adenosine monophosphate (cAMP)/protein kinase A (PKA) response element (CRE/CREB). This viral vector system offers several key advantages for molecular biology research. First, the lentiviral vector backbone efficiently and stably transduces both dividing and non-dividing cells, including primary cells that are typically difficult to transfect. Second, the CRE/CREB response element provides high specificity for monitoring cAMP/PKA signaling pathway activity, while the minimal promoter reduces background noise. Third, the firefly luciferase reporter gene allows for sensitive quantitative measurement of transcriptional activity through bioluminescence detection. The self-inactivating (SIN) design enhances biosafety by preventing viral replication, and the puromycin resistance marker facilitates the selection of stable cell lines.

This reporter lentivirus has broad applications in pharmacological and signal transduction research. It is an ideal tool for screening compounds that modulate cAMP levels or PKA activity, and is of significant value in the development of new drugs targeting G protein-coupled receptors (GPCRs) and other cAMP-related pathways. Researchers can utilize this system to study CREB-mediated gene regulation in neurological disorders, endocrine research, and metabolic disorders. The lentiviral vector format allows for the creation of stable reporter cell lines for long-term monitoring of the cAMP/PKA pathway, which is particularly useful in chronic disease models. Furthermore, it facilitates high-throughput screening of potential therapies for diseases associated with cAMP dysregulation, such as heart failure, depression, and circadian rhythm disorders.
