Arabidopsis Thaliana Transformation Service

Arabidopsis thaliana, a member of the Brassicaceae family, is used as an ideal model for molecular and physiological studies because of its short life cycle, small size, small genome (135 Mbp), ease in mutagenesis analysis and genetic transformation. Agrobacterium-mediated floral dip method with easily operating and high successful rate is widely used in Arabidopsis transformation. The general steps for this method include:

  • Target gene cloning
  • Constructing expression vector
  • Transforming plasmid into the bacterium Agrobacterium tumefaciens.
  • Growing the transformed Agrobacterium culture.
  • Dipping the Arabidopsis' flowers in Agrobacterium culture to allow for T-DNA insertions into genome.
  • Selecting for seeds that have the T-DNA insertions (usually via seed growth on antibiotic-containing media).
  • Screening for pure Arabidopsis lines in the next generation.

  • Ecotype: Col, Ws, Ler.
  • Transgenic events: at least 10 independent events.
  • Time line: 4 to 5 months from receiving construct to sending T1 seeds, 6 to 7 months from receiving construct to sending homozygous T2 seeds.
  • Capacity: on average we transform 900 constructs each year.
  • Equipment: air shower, clean bench, Bio-Rad PDS-1000/He, fluorescent microscopy, tissue culture room, greenhouse.
