Plasmids are small, circular double-stranded DNA used for manipulating and decoding genetic information in molecular biology. They have evolved into a key component of any cloning and biotechnology, as they are easier to operate. When a plasmid is introduced into the host organism through transformation, it will be replicated, resulting in many copies of the studied DNA fragment.
Plasmid purification is a technology used to separate and purify plasmid DNA from genomic DNA, protein, ribosome and bacterial cell wall. This is important for many downstream uses of plasmids, such as restricted digestion, sequencing, and transformation. On a laboratory scale, a method for isolating plasmid DNA from crude cell lysates has been well established. This plasmid DNA purification process includes the following steps: cell lysis using lysozyme, RNA removal through RNase, extraction and precipitation using organic solvents, and ultracentrifugation in a density gradient. Due to their initial design, they are very time consuming and not scalable.
