Nipah virus (NiV) is an RNA virus that belongs to the family Paramyxoviridae. Bats are the natural host of henipavirus. NiV is highly pathogenic to a variety of mammals and is considered to have pandemic potential due to its zoonotic and human-to-human transmission. In the early stages, NiV infection often manifests as febrile encephalitis or pneumonia and is difficult to distinguish from other febrile illnesses. Depending on the severity, patients may also experience fever, malaise, headache, myalgia, nausea, vomiting, dizziness, and disorientation.
The NiV genome consists of approximately 18.2 kb of single-stranded negative-sense RNA. NiV has an unsegmented negative-strand RNA genome composed of a helical nucleocapsid wrapped in an envelope, forming spherical to filamentous, pleomorphic virus particles. The genome encodes six structural proteins, namely nucleocapsid protein (N), phosphoprotein (P), matrix protein (M), fusion protein (F), glycoprotein (G) and large protein (L) or RNA polymerase, in the order 3′-N-P-M-F-G-L-5′. The three predicted nonstructural proteins, C, V, and W, are all encoded by the P gene.
