MERS-CoV ORF Clones
Middle East respiratory syndrome coronavirus (MERS-CoV) is a life-threatening human virus that emerged in Saudi Arabia in the summer of 2012. MERS-CoV has a zoonotic origin, with camels serving as intermediate hosts that transmit the virus to humans. In general, MERS-CoV infection in healthy individuals results in subclinical or mild disease. In contrast, in patients with comorbidities, MERS-CoV infection often causes acute respiratory distress syndrome and multiorgan failure, leading to death. Understanding the pathogenesis of MERS-CoV requires knowledge of the viral genes involved in virulence as well as the host cell pathways modified by the infection.
MERS-CoV belongs to the subgenus Merbecovirus of the genus Betacoronavirus. The positive-sense single-stranded RNA genome of MERS-CoV is approximately 30 kb and contains 11 open reading frames (ORFs) in the order 5'-ORF1a, ORF1b, S, 3, 4a, 4b, 5, E, M, N, 8b -3', expressed by a nested set of eight mRNAs. Genus-specific genes 3, 4a, 4b and 5 are not essential for viral replication. MERS-CoV ORF clones have invaluable potential for virological research.
