Hepatitis C Virus ORF Clones
Hepatitis C virus (HCV), a positive-strand RNA virus of the Flaviviridae family Hepacivirus, is an important blood-borne pathogen. HCV infects an estimated 2% to 3% of the world's population and is the leading cause of hepatitis, cirrhosis, and hepatocellular carcinoma. The genome consists of 5' and 3' untranslated regions (UTRs) and a single open reading frame (ORF), encoding structural proteins (Core, E1, and E2), p7, and nonstructural proteins (NS2, NS3, NS4A, NS4B, NS5A, and NS5B). HCV RNA is approximately 9600 nucleotides (nt) long and encodes a large polyprotein within a single ORF of approximately 3010 amino acids (aa). In the viral genome, the HCV ORF is preceded by a 341 nt 5' UTR, which is highly conserved among viral isolates and serves as an internal ribosome entry site to direct translation of the HCV polyprotein.
Because the different HCV genotypes differ in their biology and susceptibility to therapeutic and neutralizing antibodies, it is important to create research tools for all major genotypes.
