Large construction plasmids are plasmids designed to carry and deliver large DNA fragments or insertions, typically up to 30 kilobases (kb) or more. There are several types of large DNA-based vectors in molecular biology used for cloning and manipulating large chunks of DNA. Some of the most commonly used vectors include bacterial Artificial Chromosomes (BACs), yeast Artificial Chromosomes (YACs), cosmids and artificial chromosomes. Large Construct Plasmids can be modified to contain specific DNA sequences, such as genes, promoters, and regulatory elements, allowing researchers to manipulate and analyze the functions of these sequences in living cells or organisms. These plasmids are commonly used in molecular biology research and genetic engineering to create transgenic or genetically modified organisms.
However, large construct plasmids are usually much larger than other plasmids, which makes purification more challenging. The larger size of the plasmid can lead to an increase in physical stress during the purification process, leading to the shearing or breaking of the plasmid DNA. During the purification process, large construction plasmids may be more difficult to separate from other DNA molecules and cellular components. This may lead to contamination by other nucleic acids or proteins, thereby affecting the purity and quality of the final product. The low copy number of large plasmids in cells can lead to a decrease in the yield of purified DNA, making obtaining sufficient DNA for downstream applications more challenging.
