Rat ORF Clones
A comprehensive understanding of cellular physiology requires protein expression for protein interaction screening, cellular function screening, knockout and knockdown phenotypic validation, and many other approaches. Performing such studies at the single protein or proteome scale requires comprehensive collections of human protein-expressing clones. The open reading frame (ORF) refers to the portion of a gene's reading frame that contains the potential to encode a protein. This fragment begins with a start codon and ends with one of three stop codons. Each codon in this structure is a "read" group of three DNA or RNA nucleotide bases, indicating the formation of a certain amino acid. Thus, strings of codons will design specific protein chains. Essentially, ORF cloning is a tool for understanding gene function by elucidating the proteins produced by the gene.
The rat model was sequenced in 2004, making it the third mammalian genome to be deciphered, after humans and mice. Since their completion, rat ORF clones have gained enormous prominence, and their genetic makeup is very similar to that of humans, meaning they are the best model for understanding human disease. Using rat ORF clones, researchers can study the effects of altered gene expression. By genetically manipulating ORFs in cells derived from rat models, the scientific community can identify key genetic contributions to a variety of diseases, accelerate drug discovery, and lay the foundation for personalized medicine.
