DNA methylation is an epigenetic mechanism used by cells to control gene expression. In eukaryotes, there are many mechanisms that control gene expression, but DNA methylation is a common epigenetic signaling tool that can fix genes in the "off" position. DNA methylation is catalyzed by a DNA methyltransferase family (Dnmts), which transfers a methyl group from S-adenine methionine (SAM) to the fifth carbon of cytosine residue to form 5mC. In recent decades, scientists have made various discoveries about DNA methylation and its importance to many cellular processes such as embryonic development, X-chromosome inactivation, genomic imprinting, gene inhibition, carcinogenesis and chromosome stability. Researchers have linked abnormal DNA methylation with several adverse consequences, including human diseases.
5-methylcytosine (5-mC) is a modified version of the cytosine base (C) found in DNA. In 5-mC, a methyl group (-CH3) is added to the carbon 5 position of the cytosine ring structure. This modification can affect the structure and function of DNA, as well as gene expression, by regulating its accessibility to transcription factors and other proteins. Over the years, techniques for measuring 5-mC have been developed, whether based on heat or chemistry. Bisulfite conversion is a commonly used technique for preparing DNA for gene specific horizontal methylation analysis. This process converts cytosine into uracil while preserving the integrity of 5-methylcytosine for single nucleotide resolution methylation analysis.
