HIV ORF Clones

As the causative agent of AIDS, human immunodeficiency virus (HIV) poses a threat to global public health and economy. The HIV pandemic is characterized by extensive genomic diversity caused by multiple factors, including multiple zoonotic transmissions into the human population, high viral evolution and recombination rates. HIV isolates are currently divided into two types: HIV type 1 (HIV-1) and HIV type 2 (HIV-2). The worldwide main agent of AIDS is HIV-1, while HIV-2 is restricted to some regions of Western and Central Africa.

HIV-1 consists of two noncovalently linked, unspliced, positive-sense single-stranded RNA copies. Both copies of the RNA strand are essential for HIV-1 recombination, which occurs during reverse transcription of viral replication. The virion contains two copies of single-stranded RNA, but only a single DNA provirus is produced, called a pseudodiploid. The RNA component is 9749 nucleotides long, has a 5' cap (Gppp), a 3' poly(A) tail, and numerous open reading frames (ORFs). Viral structural proteins are encoded by long ORFs, while smaller ORFs encode regulators of the viral life cycle: attachment, membrane fusion, replication, and assembly.
