Description Human Adenovirus Type5 (dE1/E3) expressing mRed fluorescent protein (mRFP) under the control of CMV promoter. mRed fluorescent protein is the mutant Red fluorescent protein from Discosoma (DsRed), mRFP overcome the tetramerization which is toxic to cells. mRFP can be used as a control in target cells.

Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots.
Endotoxin Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance.
Sterility Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination.
Ad5 E1 Detection All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination.
RCA Assays Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources.
PFU Titering All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells.

The mRFP adenovirus is a powerful tool for molecular biology and cell research. It is based on the human adenovirus type 5 backbone with gene deletions in the E1 and E3 regions (dE1/E3). This recombinant adenovirus expresses the mRed fluorescent protein (mRFP), a monomeric variant of the DsRed protein derived from the Discosoma coral. Unlike its tetrameric precursor, mRFP overcomes the inherent toxicity associated with tetramerization, making it safer and more reliable for cellular applications. The mRFP gene is driven by a strong cytomegalovirus (CMV) promoter, ensuring robust and sustained expression in a variety of mammalian cells. Due to the deletion of the E1 gene, this adenoviral vector is replication-deficient, minimizing unintended replication in host cells while still achieving efficient transduction. Its high infectivity, broad host range, and ability to deliver genes without integration into the host genome make it ideal for transient expression studies. Furthermore, the bright red fluorescence of mRFP provides a clearly visible signal, facilitating real-time tracking of infected cells and enabling co-localization studies with other fluorescent markers.

The mRFP adenovirus is widely used as a general control vector in experimental workflows, particularly in gene delivery and cell targeting studies. Its primary role is to serve as a benchmark for transduction efficiency, allowing researchers to standardize data and validate the performance of other experimental adenoviral constructs. In target cell validation, mRFP expression serves as a visual indicator of successful infection, helping to confirm cell type specificity and infection kinetics without interfering with downstream assays. Additionally, it supports toxicity and cell viability assessments by providing an interference-free fluorescent readout, enabling long-term tracking of post-transduction cellular behavior. In live-cell imaging, the photostability of mRFP allows for prolonged observation of cellular dynamics, which is crucial for studies of migration, proliferation, and apoptosis.
