| Description | JVM2-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The expression and/or function of the Cas9 nuclease in JVM2-Cas9 cell line has been validated. In combination with separately transfected sgRNAs, JVM2-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or In pools. |
| Introduction | Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR). |

The Cas9 Stable Cell Line - JVM2 is a high-performance genetic research tool specifically engineered for streamlined genome editing in a human B-cell chronic lymphocytic leukemia (CLL) background. This line is established through the stable integration of the Streptococcus pyogenes Cas9 (SpCas9) gene into the genome of the JVM-2 host cell via lentiviral transduction. To ensure long-term stability and high expression levels, the cell line is typically maintained under selective pressure using antibiotics like Puromycin or Blasticidin. By providing a constant source of the Cas9 nuclease, this model eliminates the inconsistencies often associated with transient transfection, such as fluctuating protein levels and cellular toxicity. The JVM-2 background is particularly valuable for its well-defined phenotypic characteristics, making this stable line a reliable foundation for studying the molecular mechanisms of hematological cancers and B-cell signaling.
This cell line is extensively utilized in a variety of functional genomics applications, most notably for high-efficiency gene knockout and knock-in experiments. Given that suspension B-cells are traditionally difficult to transfect, the pre-integrated Cas9 significantly simplifies the CRISPR workflow; researchers only need to introduce the target-specific single-guide RNA (sgRNA) to achieve high editing rates. This makes the Cas9-JVM2 line an optimal choice for large-scale, high-throughput CRISPR screening aimed at identifying oncogenic drivers or uncovering novel drug resistance genes in CLL models. Additionally, the line facilitates the validation of therapeutic targets and the study of gene-drug interactions, providing a consistent biological context for testing small molecule inhibitors. Its versatility extends to complex genomic manipulations, including the creation of reporter lines or the modeling of chromosomal translocations, thereby accelerating the development of personalized medicine and targeted leukemia therapies.
