| Description | LoVo-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The Cas9 nuclease in LoVo-Cas9 cell line has been functionally validated using T7 Endonuclease I assay. In combination with separately transfected sgRNAs, LoVo-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or in pools. |
| Introduction | Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR). |

The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

The LoVo cell line was originally established in 1971 from a metastatic tumor nodule resected from the left supraclavicular region of a 56-year-old male patient diagnosed with grade IV Dukes C colorectal adenocarcinoma. Derived from the large intestine, these epithelial cells grow adherently and are known for producing carcinoembryonic antigen (CEA). The line exhibits a hyperdiploid karyotype with stable yet homogeneous chromosomal features and expresses several oncogenes, including c-myc, K-ras, H-ras, and N-ras. It is tumorigenic in nude mice. The Cas9 Stable Cell Line - LoVo is derived from this parental line through stable integration and constitutive expression of the CRISPR-associated protein 9 (Cas9) nuclease. This engineered variant eliminates the need for transient Cas9 delivery via plasmid transfection or ribonucleoprotein complexes, providing a consistent and reproducible platform for genome editing experiments. The constitutively expressed Cas9 can be readily directed by synthetic guide RNAs to induce double-strand breaks at target genomic loci.
The Cas9 Stable Cell Line - LoVo is particularly suited for loss-of-function studies in colorectal cancer research. Researchers can use it to perform high-throughput genetic screens, investigate drug resistance mechanisms, and study tumor suppressor gene function in a physiologically relevant colon adenocarcinoma background. The stable Cas9 expression streamlines experimental workflows, reducing batch-to-batch variability and allowing for more consistent editing efficiencies across multiple rounds of transfection. This cell line also supports the generation of isogenic knockout clones for downstream functional validation, including proliferation assays, apoptosis studies, and migration or invasion analyses. Additionally, it can be used in combination with synthetic guide RNA libraries for pooled CRISPR screens aimed at identifying novel therapeutic targets or genes involved in metastatic progression. The LoVo background, with its well-characterized mutational profile and CEA production, makes this Cas9-stable line a valuable tool for both basic cancer biology research and preclinical drug development efforts targeting colorectal cancer pathways.

Using the Cas9 LoVo stable cell line completely removed the bottleneck of co-transfecting Cas9 plasmids in our colon cancer studies. The constitutive Cas9 activity is excellent, and cell viability remains high post-transduction.
