| Description | HL-60-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The expression and/or function of the Cas9 nuclease in HL-60-Cas9 cell line has been validated. In combination with separately transfected sgRNAs, HL-60-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or In pools. |
| Introduction | Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR). |

The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

The HL-60 Cas9 stable cell line is a specialized derivative of the human promyelocytic leukemia cell line, originally established from a patient with acute promyelocytic leukemia. HL-60 cells are widely recognized for their ability to differentiate into mature myeloid cells, such as neutrophils or macrophages, when treated with specific inducing agents like DMSO or retinoic acid. This stable cell line has been engineered to constitutively and robustly express the Streptococcus pyogenes Cas9 (SpCas9) nuclease, typically integrated into the genome via lentiviral transduction. By maintaining high levels of Cas9 expression under a strong constitutive promoter, these cells provide a consistent genetic background that significantly simplifies the CRISPR/Cas9 workflow. Researchers can bypass the common challenges associated with transient Cas9 delivery, such as low transfection efficiency in suspension cells or cellular toxicity, ensuring more uniform and reproducible gene editing across the entire cell population.
The utility of the Cas9-expressing HL-60 cell line spans a broad range of functional genomics and biomedical research applications. It is particularly effective for large-scale loss-of-function screens using lentiviral gRNA libraries to identify critical genes involved in leukemogenesis, cell cycle regulation, and hematopoietic differentiation. Given the unique capacity of HL-60 cells to mimic primary myeloid cells, this line serves as an essential tool for studying inflammatory signaling, phagocytosis, and the mechanisms of innate immune responses. Furthermore, it enables high-efficiency gene knockout and precise knock-in experiments to model disease-associated mutations or to validate therapeutic targets in blood-borne cancers. The streamlined approach offered by these stable cells also facilitates the investigation of drug resistance mechanisms and the high-throughput screening of small-molecule inhibitors, providing a reliable platform for accelerating drug discovery and understanding the molecular basis of hematological malignancies.
