Cat. No. CSC-RO01190
Description JIMT-1-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The Cas9 nuclease in JIMT-1-Cas9 cell line has been functionally validated using T7 Endonuclease I assay. In combination with separately transfected sgRNAs, JIMT-1-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or in pools.
Introduction Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR).

The JIMT-1 cell line is a human breast cancer cell line originally derived from the pleural effusion of a 62-year-old female patient with metastatic breast cancer who exhibited resistance to trastuzumab (Herceptin). Characterized by HER2 gene amplification and overexpression, this cell line serves as an important model for studying HER2-targeted therapy resistance mechanisms. The Cas9 Stable Cell Line - JIMT-1 is a genetically engineered derivative of this parent cell line, stably expressing the Streptococcus pyogenes Cas9 endonuclease. This modification allows for stable and efficient genome editing without repeated transfections.

The Cas9 Stable Cell Line - JIMT-1 is a powerful tool in oncology and drug development. Its primary applications lie in efficient gene knockout studies, targeting the HER2 signaling pathway, tumor suppressor genes, or drug resistance mechanisms. Researchers can rapidly construct homologous cell models by introducing specific sgRNAs to study gene function, validate therapeutic targets, or simulate drug resistance phenotypes. This cell line is particularly suitable for CRISPR screening (including array screening and hybrid screening) to identify trastuzumab-sensitive genetic modifiers or synthetic lethal interactions. Furthermore, it facilitates precise gene knock-in experiments for labeling proteins or introducing patient-derived mutations. In drug development, this cell line can be used for target validation studies of novel HER2 inhibitors and combination therapies. Its stable Cas9 expression ensures consistent editing efficiency in experiments, thereby reducing variability in functional genomics studies, biomarker discovery, and breast cancer progression mechanism research.
