Description JeKo-1-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The expression and/or function of the Cas9 nuclease in JeKo-1-Cas9 cell line has been validated. In combination with separately transfected sgRNAs, JeKo-1-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or In pools.
Introduction Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR).

Safety Considerations

The JeKo-1-Cas9 stable cell line is an important immunological model established by integrating the Cas9 nuclease into the JeKo-1 mantle cell lymphoma (MCL) background. The JeKo-1 cell line was initially isolated from peripheral blood mononuclear cells of a 78-year-old female patient with large cell MCL who had undergone leukemic transformation. These cells exhibited a typical B-cell phenotype (IgM+, CD5+, CD19+) and were characterized by the t(11;14)(q13;q32) translocation, which leads to the hallmark overexpression of cyclin D1. The stable expression of Cas9 in this cell line provides a highly reproducible platform for genome editing, overcoming the challenge of low transfection efficiency commonly found in suspension-cultured lymphocytes. This system enables precise and scalable manipulation of the MCL genome, thereby facilitating research on B-cell signaling and the identification of genetic vulnerability in aggressive lymphomas.

In research, the JeKo-1-Cas9 stable cell line is primarily used for drug development and research into the molecular pathogenesis of mantle cell lymphoma. It is a powerful CRISPR gene-editing screening tool that can be used to identify genes involved in cell cycle regulation, apoptosis, and resistance to B-cell receptor (BCR) pathway inhibitors or Bcl-2 antagonists.
