| Description | KYSE-150-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The Cas9 nuclease in KYSE-150-Cas9 cell line has been functionally validated using T7 Endonuclease I assay. In combination with separately transfected sgRNAs, KYSE-150-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or in pools. |
| Introduction | Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR). |

The KYSE-150 cell line, derived from human esophageal squamous cell carcinoma, is an important model for studying esophageal cancer biology. The Cas9 Stable Cell Line - KYSE-150 is a genetically engineered variant in which the Cas9 endonuclease is stably integrated into the KYSE-150 genome. This modification ensures stable, long-term expression of the Cas9 protein without the need for repeated transfection. Its key characteristics include robust Cas9 activity, compatibility with standard cell culture protocols, and preservation of parental cell line characteristics relevant to cancer research. This engineered cell line eliminates the variability introduced by transient transfection methods, providing researchers with a reliable platform for precise gene manipulation.
The Cas9 Stable Cell Line - KYSE-150 can be applied in oncology and functional genomics. This cell line is primarily used for efficient gene knockout studies to explore tumor suppressor genes, oncogenes, and signaling pathways involved in esophageal cancer development. Researchers use this cell line for genome-wide CRISPR screening to identify novel therapeutic targets or resistance mechanisms in the context of chemotherapy and radiotherapy. Furthermore, this cell line supports precise gene knock-in experiments to introduce point mutations, tag or reporter gene constructs to mimic cancer-related gene alterations. This cell line is of immeasurable value for in vitro and in vivo studies exploring tumor progression, metastasis, and drug sensitivity.
