| Description | Daudi-Cas9 cell line is engineered to stably overexpress Cas9 nuclease. The expression and/or function of the Cas9 nuclease in Daudi-Cas9 cell line has been validated. In combination with separately transfected sgRNAs, Daudi-Cas9 cell line can be used to efficiently generate targeted genomic modifications including gene knockout, gene knockin, gene mutagenesis, gene tagging etc. It is also an ideal cell line model for sgRNA screening and validation, either individually or In pools. |
| Introduction | Clustered regularly interspaced palindromic repeats (CRISPR)/Cas9 is a gene-editing technology that contains two essential components: a guide RNA (gRNA) to match a target gene, and the Cas9 (CRISPR-associated protein 9) endonuclease which causes a double-stranded DNA break, allowing modifications to the genome via nonhomologous end joining (NHEJ) or homology-directed repair (HDR). |

The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

The Daudi Cas9 stable cell line is a specialized derivative of the well-characterized Daudi human B-lymphoblastoid line, which was originally established from a patient suffering from Burkitt’s lymphoma. This cell line is widely recognized in the scientific community for its lack of surface beta-2-microglobulin and its high expression of B-cell markers like CD19 and CD20. To create the stable Cas9 variant, the Streptococcus pyogenes Cas9 nuclease is integrated into the host genome via lentiviral transduction, typically paired with an antibiotic selection marker like puromycin to ensure population purity. This modification ensures a high and consistent level of Cas9 expression throughout the cell population over multiple passages. By using this pre-engineered line, researchers can bypass the toxicity and low efficiency often associated with the transient transfection of large Cas9-containing plasmids, establishing a streamlined, “ready-to-use” system for precise genetic manipulation.
In terms of application, the Cas9-expressing Daudi cell line is a powerful tool for functional genomics, specifically for studying B-cell malignancies and immune system interactions. It is particularly well-suited for high-throughput loss-of-function screens using sgRNA libraries, allowing scientists to pinpoint genes that drive tumor growth or confer resistance to chemotherapy. Beyond screening, the line facilitates the rapid generation of specific gene knockouts or knock-ins to investigate B-cell receptor signaling and intracellular pathways. Because Daudi cells are a standard target for antibody-dependent cellular cytotoxicity (ADCC) assays, this stable line is also frequently utilized in immunotherapy research to evaluate the killing efficiency of CAR-T cells or therapeutic antibodies. By simplifying the CRISPR workflow to the delivery of guide RNAs alone, the Daudi Cas9 stable cell line significantly enhances the speed and reproducibility of molecular research in hematology and oncology.
