Tyk2 Gene Editing    

Non-receptor tyrosine kinase 2 (TYK2) is a member of the Janus kinase (JAK) family containing three additional members (JAK1-3). JAK binds to a variety of cytokines and growth factor receptors and undergoes self and/or cross-phosphorylation upon ligand binding. Activated JAK phosphorylates members of the signal transduction and activators of transcription (STAT) family and activates several cytokines and type I interferons (IFNs). Structurally, TYK2 shares a conserved structure of seven JAK homology (JH) domains with other JAKs, wherein the C-terminal JH1 and JH2 encode a kinase and a pseudokinase domain, respectively, and the N-terminal portion provides a protein-protein interaction domain. TYK2 kinase activity is essential for undisturbed signaling, which does not exert inhibition while losing kinase activity. And more importantly, TYK2 protein stability is dependent on JH1-mediated kinase activity, which is very beneficial in the future development of Tyk2 inhibitors as a target therapeutic for human diseases.

Studies have shown that inhibition of TYK2 inhibits double-stranded RNA-induced apoptosis in human pancreatic cancer β-cells by reducing type I IFN-STAT signaling and preventing the increased expression of major histocompatibility complex (MHC)-like proteins. And there is a link between TYK2 gene variants and autoimmune diseases (such as systemic lupus erythematosus, multiple sclerosis, rheumatoid arthritis, and type 1 diabetes, etc.), inflammatory diseases, and tumors. Moreover, the function of TYK2 has been extended from effective tumor surveillance to true oncogenes. Therefore, TYK2 is an attractive therapeutic intervention target. Future work will need to further describe the structure-function relationship and fully understand the involvement of TYK2 in the immune regulation network.
