CRISPR Knockout Screening

CRISPRi Screening
CRISPR interference is developed on CRISPR/Cas9 technology. A repressor is linked to inactive Cas9 protein resulting in the repression of specific target gene. CRISPRi screening is used to study gene function by downregulating its expression and function other than completely knockout. This screening is ideal method to evaluate the function of genes that induce lethal when knockout.
CRISPRa Screening
CRISPR activation is a method that amplifies gene expression by fusing an activator to Cas9 protein. CRISPR screening provides an approach to study gain of gene function and validates loss-of-function effects from a reverse perspective. This screening helps to study the functions of genes that express low in nature condition.
