The knock-in mouse is an important research model in which an exogenous gene or DNA fragment is introduced into the mouse genome and transmitting to next generation. Knock-in mice are utilized in a wide range of application, from the study of gene function to the production of therapeutically humanized antibodies. Genome editing in mice originally relied on the random integration of a target vector in embryonic stem cells and subsequent injection of targeted clones into donor mouse blastocysts. Later, engineered nucleases were developed which make genome editing more precisely. Typical engineered nucleases include ZFNs, TALENs and CRISPR and all the three nucleases can induce double strand break in genome. Then the repair pathways in cell would be activated and finally result in either a gene knockout from NHEJ or, when a donor DNA is present, a knockin mouse from HDR. As a latest technology, CRISPR/Cas9 become a main method for genome editing mice generation.
