JAK2 is a member of the Janus kinase (JAKs), which includes JAK1-3 and TYK2. The JAK2 gene is located on the chromosome at 9p24.1. JAKs are non-receptor protein tyrosine kinases that play important roles in regulating hematopoiesis, immune system, and cellular metabolism. Binding of cytokines to the extracellular region of their cognate receptor induces receptor dimerization and promotes trans-phosphorylation (activation) of the associated JAK. Activated JAK phosphorylates specific tyrosine residues on cytokine receptors, followed by phosphorylation of signal transduction and activator of transcriptional (STAT) proteins through their SH2 (Src homology 2) domain to recruit phosphorylated receptors. The phosphorylated STAT then translocates to the nucleus to initiate a specific transcriptional program. The JAK-STAT signal transduction pathway is critical for biological development and homeostasis, especially in immunity.

JAK2 has been shown to be the predominant JAK activated in the response multiple cytokines, including Epo, IL-3, and granulocyte/macrophage colony-stimulating factor (GM-CSF), IL-5, thrombopoietin (TPO), growth hormone and prolactin. JAK2 protein is particularly important for controlling hematopoiesis in hematopoietic stem cells. These stem cells are located in the bone marrow and may develop into red blood cells, white blood cells, and platelets. Its mutations are closely related to a variety of diseases, including polycythemia vera (PV), essential thrombocythemia and myelofibrosis, and other myeloproliferative disorders. Among them, the presence of JAK2 mutation is one of the methods of clinically confirmed PV. The highly prevalent JAK2 V617F mutation was found in myeloproliferative neoplasms (MPN), and JAK2 gene fusion with TEL (ETV6) (TEL-JAK2) and PCM1 genes are also found in patients with leukemia. Therefore, JAK2 has become a major target of inhibition. Further in-depth study of JAK2 pathogenic activation will enable the development of novel small molecule targeted therapeutics.
