Fyn Gene Editing    

Fyn is a 59 kDa non-receptor tyrosine kinase belonging to the Src family kinase (SFK). The Fyn gene is located on chromosome 6q21 and expresses slightly different isomers. Among them, FynT is mainly expressed in thymocytes, spleen cells, and some blood lymphocyte lines. FynB is ubiquitous and has a particularly high expression level in the brain. The biological functions of Fyn are diverse (Figure 1). It participates in signal transduction pathways especially related to myelination in the nervous system under normal physiological conditions, as well as the development and activation of T lymphocytes. In cancer, Fyn promotes the development and progression of several cancer types by participating in the control of cell growth, death, morphogenesis, and cell movement.

One of the most well-known functions of Fyn kinase is the phosphorylation of SLAM (signaling lymphocyte activating molecule) in T cells, which induces signaling complexes that regulate interferon-gamma expression. FynT is an important mediator of T cell signaling and differentiation of CD4+ cells into T helper type 2 cells in rodents, which are critical for humoral immune responses. Recently, studies have shown that Fyn exists in the pathogenesis of AD, and its interaction with both Aβ and Tau renders Fyn a unique therapeutic target that addresses both of the major pathologic hallmarks of AD. Fyn also affects the blood system and binds to glycoprotein IIb-IIIa on platelets, a well-known integrin that has become a target for cardiovascular disease and stroke. In addition, increased expression and/or activation of Fyn is observed in various cancers, including melanoma, glioblastoma, squamous cell carcinoma, prostate cancer, and breast cancer. Studies have demonstrated the importance of Fyn in the resistance or susceptibility of cancer cells to certain anticancer therapies.
