CRISPR Rabbits for AIDS Research

AIDS has become one of the most devastating epidemics in history. An important requirement for the development of a vaccine against the AIDS pathogen human immunodeficiency virus (HIV-1) is the establishment of an easily accessible animal model to evaluate possible immunogens. To date, the only species that has been infected with HIV-1 is the chimpanzee. However, the scarcity of this animal and its designation as an endangered species severely limits its use as an animal model. Rabbits can be infected with HIV-1. Immunocompetent, permissive gene-edited rabbit models are valuable for studying HIV-1 pathogenesis and testing drug and vaccine candidates.

Solution

CRISPR/Cas9 PlatformCB optimizes rabbits with CRISPR/Cas9 technology to create immunocompetent animal models of AIDS. We developed a technique to infect genetically edited rabbits with HIV, providing researchers with a laboratory animal to test drugs and vaccines to fight this deadly disease. Within 4 weeks of being injected with HIV-infected human cells, the rabbits' immune systems began to show signs of a response to the virus, including enlarged lymph glands, spleens, and thymuses, and a decline in the number of healthy T cells, one of the cells of the immune system. We are committed to helping you understand how innate immune factors function in a species-specific environment, thereby identifying specific signaling pathways and motifs that are relevant to elucidating the mode of action of immune factors and how viruses antagonize them.

Advantages of Our Technology

Creative Biogene's CRISPR/Cas9 genome editing not only generates knockout rabbits, but also knock-in rabbits when donor ssDNA is added to gRNA and Cas9 mRNA or Cas9 protein during microinjection into embryos by gene integration into specific DSB sites in the host DNA. With the improvement of our genome editing technology, the integration rate of kb-based ssDNA has improved. Our knock-in efficiency is more than 10 times that of conventional prokaryotic microinjection.
