CDK7 Gene Editing    

CDK7 (cyclin-dependent kinase 7) is a member of the cyclin-dependent protein kinase family of serine/threonine kinases that are severely involved in the regulation of transitions between different cell cycle phases. CDK7 is thought to be a direct link between transcriptional regulation and the cell cycle.

CDK7 forms a CDK-activated kinase (CAK) complex with cyclin H and MAT1, and then phosphorylates the T-loop of CDKs in the activated segment, thereby activating CDKs to control the eukaryotic cell cycle. Cyclin H levels and CDK7 activity are unchanged throughout the cell cycle, unlike other CDKs. In addition, CDK7 is also an important component of the transcription factor TFIIH and is capable of phosphorylating the largest subunit of Pol II, the C-terminal domain (CTD), thereby playing an important role in regulating mRNA synthesis at the initial stage and DNA repair (Figure 1).

Due to the important regulatory role of CDK7 in cell division cycle and transcription, CDK7 has also become a target for small molecule inhibitors and is expected to be used for the treatment of cancer and inflammation. The current challenge is to identify relevant targets for CDK7 at each step of the transcription cycle and to understand how high dependence on an important CDK in cancer occurs and how it can be used therapeutically.
