Bcr Gene Editing    

Bcr (breakpoint cluster region protein), also named renal carcinoma antigen NY-REN-26, has serine/threonine kinase activity and is a GTPase activating protein of p21Rac. Bcr contains a unique structure for two opposite regulatory activities of small GTP-binding proteins and is associated with the signal transduction of Rho GTPases and the regulation of Rac1 activity. Its C-terminus is a GTPase activating protein (GAP) domain that stimulates GTP hydrolysis by Rac1, Rac2, and CDC42, accelerates the intrinsic rate of GTP hydrolysis of Rac1 or CDC42, resulting in downregulation of the active GTP binding form. The central Dbl homology (DH) domain acts to regulate the GTPase CDC42, RhoA and Rac1 guanine nucleotide exchange factor (GEF), promoting the conversion of CDC42, RhoA, and Rac1 from GDP binding to GTP binding. The amino terminus contains intrinsic kinase activity.

Studies have shown that Bcr can regulate macrophage function by regulating Rac1 activity and play an important negative regulatory role in neuronal Rac1 activity. Moreover, the BCR-ABL oncoprotein oligomerization domain found at the N-terminus of BCR is essential for the carcinogenicity of the BCR-ABL fusion protein. BCR-related diseases include leukemia, chronic granulocytes and leukemia, and acute lymphocytes. At present, it is unclear about the normal Bcr function and it is still unclear. Further researches are needed to help understand the relationship between Bcr and disease development.
