| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

| Background | Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008] |
EF1a-hTERT (RFP, Bla) Lentiviral Particles represent a highly efficient system for the stable expression of human Telomerase Reverse Transcriptase (hTERT) in mammalian cells. This specific construct is regulated by the elongation factor 1-alpha (EF1a) promoter, a constitutive driver known for maintaining high levels of transgene expression even in sensitive cell types where other promoters, such as CMV, may undergo epigenetic silencing. To facilitate ease of use, the particles integrate a Red Fluorescent Protein (RFP) as a visual reporter for monitoring transduction efficiency and a Blasticidin resistance marker for the rapid selection of stable integrants. These third-generation, VSV-G pseudotyped lentiviral particles are engineered for high biosafety and broad tropism, enabling the delivery of the hTERT gene into a diverse array of target cells, including primary cells, stem cells, and hard-to-transfect lines, while ensuring long-term genomic stability.
The application of these lentiviral particles is primarily focused on cell immortalization, providing researchers with a reliable method to extend the proliferative capacity of primary human cells. By counteracting the natural shortening of telomeres during cell division, hTERT expression allows cells to bypass replicative senescence, making it possible to establish stable, long-lived cell models that retain their original phenotypic traits. This is particularly valuable in fields such as cancer research, where telomerase activation is a key area of study, and in regenerative medicine, where expanded populations of primary cells are needed for therapeutic development. Additionally, the dual-marker system allows for precise cell sorting and the generation of homogeneous cell lines for high-throughput drug screening and functional genomic assays, significantly streamlining the workflow for longitudinal biological studies and complex tissue engineering projects.

Used this to extend the lifespan of our cell line—consistent TERT expression over multiple passages. The dual markers (RFP/Bla) were a huge plus. Will buy again!
