Application The human TERT lentivirus (EF1α, Hygro) has a variety of applications, primarily in biological and medical research. Here are some of the key applications: Cell immortalization: It is often used to immortalize primary cells that otherwise have a limited lifespan in culture. This allows researchers to study cellular mechanisms over longer periods of time without causing the cells to age. Disease modeling: Immortalized cells can be used to create more stable and reproducible in vitro models of disease, which is critical for understanding disease pathology and drug screening. Cancer research: Since telomerase is upregulated in many cancers, introducing human TERT into cells can help researchers study the carcinogenesis process and the role of telomerase in cancer cell proliferation and survival. Drug screening and development: Immortalized cell lines are extremely valuable for high-throughput drug screening. They provide a consistent and reproducible platform for testing potential therapeutic compounds. Genetic engineering and functional studies: Researchers can use TERT-expressing cells to study the functional effects of other genetic modifications or introduce additional genetic elements to create complex models for a variety of biological processes.

Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Customer Reviews

The Human TERT Lentivirus (EF1α, Hygro) is a powerful tool designed for advanced gene delivery and expression in mammalian cells. This lentiviral vector expresses human telomerase reverse transcriptase (TERT), a key enzyme responsible for maintaining telomere length and genomic stability, under the control of the EF1α promoter. Untagged TERT ensures native functionality of the protein without any modifications that could interfere with its activity or localization. The lentiviral system itself is favored for its ability to transduce a variety of dividing and non-dividing mammalian cells, stably integrate into the host genome, and maintain long-term expression of the transgene. The EF1α promoter is known for its robust, ubiquitous, and constitutive expression in a variety of mammalian cell types, making it an ideal promoter for a variety of research applications. It ensures consistent and high levels of TERT expression, which is critical for experiments requiring sustained telomere lengthening or for studies of cellular senescence, cancer research, and stem cell biology. A notable feature of this lentiviral vector is the inclusion of an antibiotic resistance marker - hygromycin resistance. This allows for efficient selection of transduced cells, ensuring that only cells that have successfully integrated the lentiviral vector and express the hygromycin resistance gene will survive in the presence of hygromycin.
