| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

| Background | Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008] |
The EF1a-hTERT (GFP, Puro) Lentiviral Particles are a high-performance genetic delivery tool designed for stable and constitutive expression of the human Telomerase Reverse Transcriptase (hTERT) gene. This system utilizes the human Elongation Factor 1 alpha (EF1a) promoter, which is preferred for its ability to drive robust, long-term transgene expression while remaining less susceptible to transcriptional silencing than the CMV promoter, particularly in stem cells and primary cultures. These particles are engineered as a polycistronic vector containing both a Green Fluorescent Protein (GFP) reporter for visual monitoring of transduction efficiency and a Puromycin resistance marker to allow for the rapid selection of stable cell lines. As a replication-incompetent lentiviral system, it ensures high-titer delivery and genomic integration, providing a reliable method for modifying hard-to-transfect mammalian cells with minimal toxicity.
The primary application of these lentiviral particles is the immortalization of primary cell lines, allowing researchers to bypass replicative senescence and extend the proliferative capacity of finite cell populations. By maintaining telomere length through hTERT overexpression, scientists can establish consistent in vitro models that retain the phenotypic characteristics of their original tissues, which is essential for longitudinal studies in cell biology. Beyond immortalization, the GFP-Puro dual marker system makes these particles ideal for optimizing transduction protocols and enriching modified populations via fluorescence-activated cell sorting (FACS) or antibiotic selection. These tools are frequently employed in cancer research to investigate the mechanisms of telomerase activation and in drug discovery as a platform for high-throughput screening. Additionally, they serve as a critical resource in regenerative medicine for studying the aging process and the development of engineered cell therapies.
