| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

| Background | Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008] |
EF1a-hTERT(Neo) Lentiviral Particles are a sophisticated genetic tool designed for the robust and stable expression of the human telomerase reverse transcriptase (hTERT) gene in a wide variety of mammalian cells. These replication-incompetent particles utilize the human Elongation Factor 1-alpha (EF1a) promoter, which is specifically chosen for its ability to drive high-level, constitutive transgene expression while remaining resistant to the transcriptional silencing that often affects CMV promoters in stem cells and primary cultures. The vector backbone includes a Neomycin (Neo) resistance marker, which facilitates the efficient selection of stable cell populations using G418 antibiotic treatment. Produced through a high-titer manufacturing process, these lentiviral particles ensure high transduction efficiency in both dividing and non-dividing cells, integrating the hTERT expression cassette directly into the host genome for permanent genetic modification.
The primary application of these lentiviral particles is the immortalization of primary cells, allowing researchers to bypass replicative senescence and create continuous cell lines that maintain the phenotypic characteristics of their original tissue. Unlike traditional immortalization methods involving viral oncogenes, hTERT expression preserves a more natural cellular state, making these models ideal for studies in aging, oncology, and regenerative medicine. Additionally, EF1a-hTERT(Neo) particles are widely employed in the development of cell-based assays for drug discovery and toxicology, where a consistent and renewable cell source is mandatory. They also serve as a vital tool in telomere biology research, enabling the exploration of telomerase activity in cell survival, DNA repair mechanisms, and long-term proliferative capacity. By providing a reliable platform for stable hTERT overexpression, these particles support the establishment of complex, longitudinal biological models that are essential for modern biomedical research.
