| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

| Background | Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008] |
The EF1a-hTERT Lentiviral Particles are a sophisticated molecular tool engineered for the stable delivery and constitutive expression of the human telomerase reverse transcriptase (hTERT) gene. These particles leverage the Human elongation factor-1 alpha (EF1a) promoter, a potent regulatory element that ensures high-level transcriptional activity across an expansive variety of mammalian cell lines, including notoriously difficult-to-transduce primary cells and pluripotent stem cells. Unlike other viral promoters that may face epigenetic silencing over time, the EF1a promoter provides reliable, long-term expression of the hTERT protein. This system employs a third-generation lentiviral vector framework, which enhances biosafety by utilizing a split-genome packaging strategy. Upon transduction, the viral RNA is reverse-transcribed and integrated into the host cell’s genome, providing a permanent genetic modification that enables the cell to overcome telomere-dependent replicative senescence.
In terms of research utility, the EF1a-hTERT Lentiviral Particles serve as a cornerstone for the creation of immortalized yet phenotypically normal cell lines. By replenishing telomeric DNA, hTERT allows researchers to bypass the Hayflick limit in primary human cells—such as fibroblasts, endothelial cells, and specialized immune cells—without inducing the genomic instability or oncogenic transformation often associated with traditional immortalization methods like the SV40 Large T antigen. These established cell lines are invaluable for high-throughput drug discovery, complex disease modeling, and longitudinal studies of cellular signaling pathways. Moreover, the use of the EF1a promoter makes these particles ideal for experiments involving cell differentiation, as the promoter remains active throughout various developmental stages. This functionality is particularly advantageous in regenerative medicine and tissue engineering, where maintaining a stable cell population with a consistent genotype is essential for evaluating long-term biological outcomes and therapeutic efficacy.
