| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

| Background | Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008] |
The EF1a-hTERT (RFP, Puro) Lentiviral Particles represent a sophisticated tool for the stable expression of human telomerase reverse transcriptase in mammalian systems. Utilizing the elongation factor-1 alpha (EF1a) promoter, these particles provide consistent and high-level transcription, specifically in cell types where other promoters, such as CMV, may be prone to epigenetic silencing. The vector is engineered with a dual-reporter and selection system, featuring Red Fluorescent Protein (RFP) for non-invasive visualization and a Puromycin resistance gene to facilitate the generation of pure, stable cell lines. These lentiviral particles are provided in a ready-to-use, high-titer format and are typically pseudotyped with the VSV-G protein, allowing for a broad tropism that covers almost all mammalian cell varieties, including non-dividing and primary cells.
These lentiviral particles are predominantly employed in cell engineering to achieve cellular immortalization. By introducing hTERT, researchers can extend the lifespan of primary cells indefinitely while preserving their original physiological properties and karyotypic stability. This makes them indispensable for creating robust disease models, particularly in studies involving human fibroblasts, epithelial cells, or mesenchymal stem cells. Beyond immortalization, the integrated RFP marker allows for the precise monitoring of cell population dynamics and migration in complex microenvironments or co-culture systems. The system is also highly effective for creating long-term cultures required for pharmaceutical toxicology assays and vaccine development, where consistent cellular performance over multiple passages is critical. By providing a reliable method for stable gene integration, these particles streamline the workflow from initial transduction to high-throughput functional analysis.
