| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

| Background | Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008] |
EF1α-hTERT (Bla) Lentiviral Particles are designed for stable delivery and expression of human telomerase reverse transcriptase (hTERT) in mammalian cells. hTERT is the catalytic protein subunit of telomerase, which maintains telomeric DNA at chromosome ends and helps counteract the progressive telomere shortening associated with repeated cell division. In many primary and finite-lifespan cells, hTERT expression can delay replicative senescence, extend proliferative capacity, and support the establishment of continuously expanding cultures. The human EF1α promoter drives constitutive hTERT expression and is commonly used when sustained activity is required across a broad range of cell types. A blasticidin resistance gene enables antibiotic selection and enrichment of successfully transduced cells. Lentiviral delivery provides efficient gene transfer and stable genomic integration, including in cell populations that are difficult to modify by conventional plasmid transfection.
These particles are primarily used to generate hTERT-immortalized models from primary cells or other cultures with limited replicative potential. Suitable applications include the expansion of epithelial cells, endothelial cells, fibroblasts, stromal cells, and additional lineage-specific populations for long-term experimental use. hTERT-expressing cells can support studies of cellular aging, telomere biology, disease mechanisms, signal transduction, gene function, and responses to drugs, biologics, or environmental stress. They can also provide renewable starting material for assay development, compound screening, toxicology studies, biomarker analysis, and subsequent genetic engineering. Compared with repeatedly sourced primary cultures, a well-characterized hTERT-immortalized population may reduce donor-to-donor and passage-related variability and improve experimental consistency.
