Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.
Target Gene TERT
Background Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined. Alternative splicing at this locus is thought to be one mechanism of regulation of telomerase activity. [provided by RefSeq, Jul 2008]

The EF1a-hTERT (Zeo) Lentiviral Particles represent a high-titer gene delivery system designed for the stable expression of the human telomerase reverse transcriptase (hTERT) protein. These particles are engineered using a third-generation lentiviral vector system, ensuring high transduction efficiency and stable integration of the transgene into the host genome. The expression of hTERT is controlled by the human elongation factor-1 alpha (EF1a) promoter, which is preferred for its ability to maintain high-level, constitutive expression across a diverse range of cell types, including those where traditional promoters like CMV are prone to transcriptional silencing. To facilitate the selection of successfully transduced cells, the vector includes a Zeocin resistance gene, allowing researchers to generate stable, homogenous cell populations through antibiotic selection.

The primary application of EF1a-hTERT (Zeo) Lentiviral Particles is the immortalization of primary cells. By maintaining telomere length, the ectopic expression of hTERT allows primary cells to bypass replicative senescence—the Hayflick limit—enabling them to proliferate indefinitely while retaining their original phenotypic and functional characteristics. This makes the particles an essential tool for establishing long-term, reproducible cell models for rare or hard-to-culture tissues, such as human fibroblasts, epithelial cells, and mesenchymal stem cells. Furthermore, these particles are widely utilized in aging research, oncology studies, and drug toxicity screenings that require a consistent supply of non-transformed cells. They are also instrumental in tissue engineering and regenerative medicine, where maintaining the proliferative potential of progenitor cells is critical for downstream applications. By providing a reliable method for genomic integration, this product ensures a stable cellular platform for complex longitudinal studies and high-throughput biological assays.
