Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The EF1a-hKRAS/G12V (GFP, Puro) Lentiviral Particles provide a highly efficient system for the stable delivery and expression of the oncogenic human KRAS G12V mutation in mammalian cells. Driven by the constitutive EF1a promoter, the vector ensures high-level expression across various cell lines, including those typically resistant to traditional transfection or prone to promoter silencing, such as primary and stem cells. This specific KRAS variant contains a substitution at codon 12, resulting in a constitutively active GTP-bound state that drives uncontrolled cellular signaling. The particles are engineered as third-generation, replication-deficient units, incorporating a bicistronic expression cassette that includes Green Fluorescent Protein (GFP) for direct visualization and a Puromycin resistance marker for the selection of stable clones. This integration facilitates both the immediate assessment of transduction efficiency and the long-term maintenance of the transgene.

These lentiviral particles are indispensable tools for investigating the molecular drivers of oncogenesis and evaluating targeted therapeutic interventions. Researchers utilize them to establish stable cell lines that mimic the signaling environment of KRAS-driven cancers, such as pancreatic, colorectal, and lung adenocarcinomas. Beyond basic signaling research into the MAPK and PI3K pathways, these particles are extensively used in high-throughput drug screening to identify small molecules that specifically inhibit mutant KRAS or its downstream effectors. The GFP reporter enables sophisticated applications, including longitudinal live-cell imaging, flow cytometry-based population analysis, and the tracking of tumor cell migration or invasion in 3D organoid models. Furthermore, the ability to generate stable, antibiotic-selected populations makes these particles ideal for in vivo studies, including subcutaneous or orthotopic xenograft models, where consistent expression of the G12V mutant is required to observe phenotypic changes in tumor growth and metastasis.
