Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

EF1a-hCDK4(RFP, Bla) lentiviral particles provide a powerful and versatile tool for gene manipulation in mammalian cells. Modified with the EF1a promoter, this lentivirus ensures stable and sustained expression of the human CDK4 gene in a variety of cell types, including primary cells and those difficult to transfect. Its included red fluorescent protein (RFP) reporter gene allows for easy visualization and tracking of transduced cells via fluorescence microscopy or flow cytometry, enabling real-time monitoring without additional staining or fixation. Furthermore, the blast fungicide resistance gene (Bla) provides a highly efficient screening mechanism, allowing researchers to remove untransduced cells under blast fungicide treatment, thereby establishing stable cell lines. This dual-function system, combining a strong promoter, a visual reporter gene, and a selection marker, ensures high transduction efficiency, reliable gene expression, and a simplified workflow.

The primary application of EF1a-hCDK4(RFP, Bla) lentiviral particles is cell immortalization, particularly suitable for primary human cells that typically age after limited passages. By overexpressing CDK4 (a key regulator of the G1/S phase transition in the cell cycle), this lentivirus enables cells to bypass replicative senescence, thereby extending their lifespan and facilitating long-term research. Furthermore, the lentiviral particles play a crucial role in investigating the function of CDK4 in tumorigenesis, drug resistance, and cell cycle dysregulation. The RFP reporter gene allows for precise isolation of transduced cells via fluorescence-activated cell sorting (FACS), resulting in a homogeneous cell population for subsequent proteomics, transcriptomics, or high-throughput screening analyses. Beyond cell immortalization, this tool also supports co-transduction experiments with other lentiviral vectors (e.g., expressing cyclin D or other cell cycle regulators) to mimic complex signaling pathways.
