Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The EF1a-SV40/LTA (GFP, Puro) Lentiviral Particles are high-titer, replication-deficient viral vectors designed for the stable delivery and constitutive expression of the SV40 Large T Antigen (LTA). This construct is driven by the human elongation factor-1 alpha (EF1a) promoter, which is known for its high-level expression and resistance to gene silencing, making it particularly effective in primary cells, stem cells, and other challenging mammalian cell types. To facilitate experimental tracking and selection, the particles incorporate a Green Fluorescent Protein (GFP) reporter for the visual assessment of transduction efficiency and a Puromycin resistance marker for the generation of stable cell lines. Produced using a third-generation lentiviral packaging system, these particles ensure high biosafety standards and robust integration into the host genome, providing a reliable platform for permanent genetic modification.

The primary application of these lentiviral particles is the immortalization of primary cells to establish continuous cell lines. By sequestering key tumor suppressor proteins, specifically p53 and the retinoblastoma (pRb) family, the SV40 Large T Antigen allows cells to bypass replicative senescence and maintain a proliferative state. This makes it an essential tool for researchers who need to expand limited primary cell populations for long-term functional studies. Beyond immortalization, these particles are widely used in research concerning cell cycle regulation, oncogenic transformation, and viral DNA replication mechanisms. The integrated GFP and Puromycin markers streamline the experimental workflow, allowing for the rapid identification and selection of successfully transduced populations. Whether used for developing robust cellular models for drug screening or investigating the molecular pathways of cancer progression, these lentiviral particles offer a high-performance solution for long-term transgene expression.
