Knock-in Rats Model    

CRISPR/Cas9 PlatformCB offers you CRISPR-based gene knock-in rats at very competitive prices. those models are suitable for a variety of applications, including reporter gene expression, conditional gene expression, and disease models. We guarantee at least 2 founders or 3 F1 animals. Tell us the name of your gene and the target site you want to target; we will generate your rat model.

Our knock-in rats model includes

  • Fluorescent proteins or immune-tags insertion, custom tags and combinations are also available.
  • Cre gene and Floxed allele insertion (for conditionally knockout target gene).
  • Large fragment insertion at Rosa26 - Targeting Rosa26 can control gene copy number and gene expression, and avoid silencing or suppressing the expression of neighboring genes.

Project workflow

Knock-in Rats Model

  • Step1. Knock-in strategy design

Consult our team of experts and tell us what genes and loci you want to knock in. We will customize high-efficiency targeting strategies for you, including the most suitable PAM sites, donor DNA and efficient gRNA sequences design, as well as genotyping primers design based on PCR and sequencing.

  • Step2. Knock-in vector construction

Synthesize gRNA, Cas9 mRNA, donor DNA, and construct CRISPR/Cas9 vector.

  • Step3. CRISPR/Cas9 injection

gRNA, Cas9 mRNA and donor DNA are simultaneously implanted into rat single-cell embryos (as many as possible).

  • Step4. Founder screening

Select the young rats with right gene editing by PCR and sequencing.

  • Step5. Breeding founders to obtain F1 (optional)
  • Step6. Delivery

We will delivery at least 2 founders or 3 F1 rats, and final report which details each step, including target vector design, construction and validation, transfection conditions, genotyping strategies and results.
