Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv(TACSTD2)-CD3zeta CAR-T Lentivirus is a high-titer, replication-incompetent viral vector designed for the stable expression of a TROP2-specific Chimeric Antigen Receptor (CAR) in primary human T cells. The genetic payload encodes a specialized fusion protein comprising a high-affinity single-chain variable fragment (scFv) derived from an anti-TACSTD2 antibody, linked via a transmembrane hinge to the CD3-zeta (CD3ζ) intracellular signaling domain. TACSTD2, also known as Tumor-Associated Calcium Signal Transducer 2 or TROP2, is a cell-surface glycoprotein that is significantly upregulated in numerous epithelial malignancies. By utilizing a lentiviral delivery system, this product ensures efficient integration of the CAR construct into the host cell genome, allowing for the generation of effector T cells that can recognize and bind to TROP2-expressing cells in an MHC-independent manner. This binding event triggers the CD3ζ signaling cascade, leading to T-cell activation, proliferation, and the subsequent release of cytotoxic granules to induce apoptosis in target tumor cells.

This lentiviral product is an essential tool for researchers focused on the development of adoptive cell therapies for solid tumors. Given the broad expression of TROP2 in cancers such as triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and pancreatic ductal adenocarcinoma, the scFv(TACSTD2)-CD3zeta CAR-T system is primarily used in preclinical efficacy studies to evaluate the potency of redirected T cells. It is widely employed in in vitro cytotoxicity assays to measure specific lysis of cancer cell lines and in cytokine profiling experiments to monitor the secretion of IFN-gamma and IL-2 upon antigen engagement. Additionally, the lentivirus is instrumental in establishing humanized or cell-line-derived xenograft (CDX) mouse models to study T-cell infiltration, persistence, and the metabolic challenges within the tumor microenvironment. Beyond basic oncology research, it serves as a reliable benchmark for testing novel modifications in CAR architecture, such as the addition of various co-stimulatory domains or the implementation of “armored” CAR strategies to enhance therapeutic durability.
