Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

scFv(HER2)-CD3ζ CAR-T Lentivirus is a gene-delivery reagent designed to generate T cells that recognize human epidermal growth factor receptor 2 (HER2/ERBB2). The encoded chimeric antigen receptor combines an extracellular anti-HER2 single-chain variable fragment (scFv) with membrane-anchoring elements and the intracellular signaling domain of CD3ζ. The scFv provides antibody-like recognition of cell-surface HER2 without requiring peptide presentation by the major histocompatibility complex, while CD3ζ initiates T-cell activation following receptor engagement. Because this construct contains CD3ζ without an additional costimulatory domain, it represents a first-generation CAR design. Lentiviral delivery enables efficient gene transfer into dividing and non-dividing cells and supports stable CAR expression in transduced T cells, providing a consistent starting point for downstream functional studies.

This product is intended for research into HER2-directed cellular immunotherapy and the biology of antigen-specific T-cell responses. It can be used to prepare experimental CAR-T cells for assessing receptor expression, HER2-dependent activation, target-cell killing, cytokine release, proliferation, and changes in T-cell phenotype. Researchers may compare responses across HER2-high, HER2-low, and HER2-negative cell models to examine antigen density, specificity, and potential off-target activity. The resulting cells are also suitable for evaluating HER2-positive tumor models, including models derived from breast, ovarian, gastric, and other solid tumors in which HER2 expression is relevant. As a first-generation construct, scFv(HER2)-CD3ζ CAR-T Lentivirus can serve as a useful reference when studying the contribution of costimulatory domains or comparing alternative CAR architectures. It may also support preclinical investigations of combination strategies, tumor-cell resistance, immune escape, and the influence of the tumor microenvironment on CAR-T-cell function.
